Asparagine-linked glycosylation 3 (ALG3)はER膜内腔での糖鎖修飾の第一段階を担う酵素で,細胞質側で合成されたMan5GlcNAc2にマンノース残基をalpha1,3-結合で転移する機能をもつ.この酵素は糖鎖を保持する真核生物に高度に保存されているにも関わらず,植物細胞では遺伝子,及び機能同定がなされていなかった.本報ではシロイヌナズナに存在するALG3のクローニング,機能同定を行い,植物細胞内における糖鎖修飾合成経路を明らかにした.またシロイヌナズナALG3欠損変異体では野生型には存在しない糖鎖を保持するが,野生型と同様の生育を示した。ヒトでは糖鎖修飾不全が重篤な疾病となるが、植物の場合必ずしも生育に影響を及ぼさないことを明らかにした.(文責; 梶浦裕之)

N-アセチルグルコサミン転移酵素I(GnTI)は、N結合型糖鎖修飾経路において高マンノース型糖鎖から複合型糖鎖を生合成する際の鍵となる酵素である。しかしながら高等植物由来GnTIの生化学的諸性質には不明な点が多かった。本報ではタバコ由来GnTIをアフィニティタグとの融合タンパク質として大腸菌で発現し、活性を保持した状態で精製した。さらにこの精製酵素を用い、至適温度・基質特異性等の酵素学的諸性質を明らかにした。(文責;特任研究員 土肥浩二)

{alpha}-mannosidase I(MANI)は小胞体からゴルジ体に輸送されてきた糖鎖の?1,2-結合したマンノース残基を加水分解する酵素である.MANIは動物細胞をはじめとする多くの生物種で保存された酵素であるにも関わらず,植物細胞においてはクローニング,及び機能同定がされていなかった.本報ではシロイヌナズナが2種類のMANI(AtMANI)を保持することを見出し,さらに細胞内局在,酵素学的性質を明らかにした.また両遺伝子欠損変異体シロイヌナズナの糖鎖構造を解析することで,シロイヌナズナ内におけるマンノース残基の加水分解は2種類のAtMANIにより担われていることを証明した。(文責; 梶浦裕之)

グラム陰性菌リポ多糖に含まれる3-deoxy-D-manno-octulosonate (KDO)は菌の成長に必須の糖である。一方で、植物は細胞壁にKDOを持つもののその代謝経路や役割はほとんど分かっていない。本報では、植物におけるKDOの代謝・合成経路を明らかにする第一歩として、グラム陰性菌由来とは異なる酵素学的性質(至適pHなど)を持つcytidine monophosphate (CMP)-KDO合成酵素をシロイヌナズナより見出した。 (文責;助教 三崎亮)
